Abstract
Extracellular vesicles (EVs) are increasingly studied as biomarkers and potential therapeutic tools, yet how EV preparations are quantified and dosed can significantly affect experimental comparisons and interpretation.
In this study, researchers analyzed small extracellular vesicle-enriched fractions from 19 cancer and non-cancer cell lines and found that particle number and protein content provide distinct—and not interchangeable—measures of EV output. Cancer-derived cells showed approximately 3.6-fold higher estimated vesicle release per cell but approximately 4.3-fold lower protein-to-particle ratios than non-cancer cells. As a result, matching EV preparations by protein content alone can deliver substantially different particle numbers, while matching by particle number can produce equally substantial differences in protein exposure.
The findings highlight the limitations of relying on a single measure for EV normalization and dosing and support a multidimensional approach incorporating vesicle release, protein release, and protein-to-particle ratios. This framework can enable more biologically meaningful comparisons between EV preparations and improve the consistency and interpretation of EV research.
