Abstract
Poly-microbial keratitis requires rapid, broad-spectrum antimicrobial therapy, but antimicrobial resistance and dosing challenges limit current treatment options. This study developed and validated a simultaneous LC-MS/MS method to quantify amphotericin B, tobramycin, and vancomycin in rabbit ocular biofluids and tissues, then used the assay to characterize the ocular pharmacokinetics of these combination antimicrobials and predict PK-PD indices and optimal dosing frequency using Phoenix WinNonlin. The validated method showed good linearity, precision, and accuracy across relevant concentration ranges for all three drugs using minimal sample volumes. This work provides a practical bioanalytical and PK-PD framework to help optimize combination antimicrobial therapy for poly-microbial keratitis.

Phoenix WinNonlin: PK-PD for Ocular Combination Therapy
Phoenix WinNonlin predicted optimal dosing frequency for a three-drug ocular combination in this keratitis study, built on a validated simultaneous assay. Within a Phoenix platform trusted by scientists and regulators for over three decades, Phoenix WinNonlin's output can become submission-ready tables, figures, and listings through TFL Studio.
