Abstract
Microbial keratitis often requires combination therapy with antifungal, antibacterial, and anti-inflammatory agents, creating a need for methods to characterize the ocular pharmacokinetics of such combinations. This study developed and validated a simultaneous LC-MS/MS method for quantifying betamethasone, amphotericin B, azithromycin, and neomycin in rabbit tear fluid and cornea, then used single-dose ocular pharmacokinetic data and MIC values to predict optimal dosing frequency via PK-PD simulation using Phoenix WinNonlin. The validated method required only a small ocular sample volume and showed good linearity, precision, and accuracy for all four analytes. Simulation results suggested dosing frequency should be tailored to the specific pathogen and strain to minimize antimicrobial resistance in treating microbial keratitis.
