Abstract
Ivermectin, used in mass drug administration programs to eradicate lymphatic filariasis, shows high inter-individual variability in drug exposure that had not been well characterized through population pharmacokinetic modeling. This study analyzed 724 plasma samples from filariasis-infected and uninfected adults in Côte d'Ivoire following a single ivermectin dose as part of triple-drug therapy, building a population pharmacokinetic model with Phoenix NLME and simulating exposure across different dosing groups. Filarial infection status did not affect ivermectin pharmacokinetics, but sex was a significant covariate for peripheral volume of distribution, with men showing substantially lower values than women; drug exposure scaled linearly with dose regardless of sex. This first population pharmacokinetic study of ivermectin in lymphatic filariasis patients helps quantify sources of exposure variability relevant to mass drug administration programs.

Phoenix NLME:Understanding Exposure Variability
Phoenix NLME built the first population PK model for this antiparasitic in mass drug administration, isolating sex as a key driver of exposure variability. Phoenix NLME can be paired with Modeling Assistant, an AI copilot for model building and PML authoring, as part of a Phoenix platform trusted by scientists and regulators for over three decades.
