Abstract
Individual variability in response to the sedative dexmedetomidine is common, but the genetic factors underlying this variability were not well understood. This prospective cohort study of 194 female surgical patients examined associations between dexmedetomidine sedative response and genetic polymorphisms in drug-metabolizing and drug-response genes, with pharmacokinetic analysis of a subset of participants performed using Phoenix WinNonlin. A CYP2A6 polymorphism (rs28399433) was associated with lower metabolic enzyme efficiency, higher plasma dexmedetomidine concentrations, and greater sedative sensitivity, while a GABRA2 polymorphism (rs279847) was strongly associated with heart rate abnormality risk. These findings identify specific genetic variants that may help explain and predict individual variability in dexmedetomidine's sedative and cardiovascular effects.

Phoenix NLME:Genetics Behind Drug Response
Phoenix NLME's pharmacokinetic analysis supported this dexmedetomidine study, helping link two genetic variants to differences in sedative response and cardiovascular risk. As part of a Phoenix platform designed to move teams from data to decision in a fraction of the time, Phoenix NLME can be paired with Modeling Assistant, an AI copilot for model building and PML authoring.
